目的通过构建大风藤Cocculus orbiculatus根与茎的指纹图谱,对比两者相似度,为科学评价大风藤根与茎药用价值及临床应用提供参考。方法采用高效液相色谱法对贵州省14个产地20批的大风藤根与茎进行色谱分离,色谱条件为Agilent ODS C_(18)(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,运行时间为95 min,体积流量1.0 mL/min,测定波长254 nm,柱温30℃。使用“中药色谱指纹图谱相似度评价系统2.0”进行数据处理,对大风藤根与茎进行相似度分析。结果建立了大风藤根与茎指纹图谱共有模式,归纳出大风藤根与茎共有峰8个,指纹图谱相似度为0.985~0.998。结论大风藤根与茎化学成分相近,但不同产地和部位的大风藤所含活性物质的量具有一定差异。
OBJECTIVE: To establish the fingerprint of root and stem of Cocculus orbiculatus and compare the similarity between the two, provide a reference for the scientific evaluation of the medicinal value and clinical application of the root and stem of Calendula officinalis. Methods High performance liquid chromatography (HPLC) was used to separate the roots and stems of 20 batches of Gynura divaricata in 14 producing areas in Guizhou Province. The chromatographic conditions were Agilent ODS C 18 (250 mm × 4.6 mm, 5 μm) column. The mobile phase consisted of acetonitrile- 0.1% phosphoric acid aqueous solution with gradient elution. The running time was 95 min, the volume flow rate was 1.0 mL / min, the detection wavelength was 254 nm and the column temperature was 30 ℃. Using “Chinese medicine chromatographic fingerprinting similarity evaluation system 2.0 ” data processing, the similarity analysis of the root and stem of the gale. Results The common pattern of root and stem fingerprints of Gynostemma pentaphyllum was established. There were 8 peaks between roots and stems of Radix Gynostemmae, and the similarity of fingerprints was 0.985-0.998. Conclusion The chemical constituents of roots and stems of Gynura divaricata are similar, but there are some differences in the contents of active substances contained in Gynura divaricata from different habitats and locations.
