目的:探讨大鼠L5神经根受压后,益气化瘀方在神经再生修复过程中对神经肌肉接头部神经元的作用和表现。方法:采用多克隆蛋白基因产物9.5(proteingeneproduct9.5,PGP9.5)作为神经元标记,大鼠L5神经根受压后,给予益气化瘀方灌胃,分别于术后10,20,30和60d取比目鱼肌,用免疫组织化学方法结合激光共聚焦扫描显微技术,观察神经肌肉接头部末梢神经再生修复过程。α-bungarotoxin(α-BTX)荧光结合剂显示运动终板,NIH图像分析技术测定末梢神经与运动终板重叠面积。结果:肌肉失神经支配后,益气化瘀方组的末梢神经在神经肌肉接头部的聚集,出芽及延伸均明显早于对照组。神经再生修复期间,末梢神经与运动终板的重叠速度和范围以及神经肌肉接头的再构筑,益气化瘀方组亦同样明显优于对照组。结论:益气化瘀方能促进神经元的增生及提高其再生功能,加快神经肌肉接头的重建及明显缩短神经再生修复的进程。
OBJECTIVE: To investigate the effect and mechanism of Yiqi Huayu Decoction on nerve neuromuscular junction neurons in the process of nerve regeneration after rat L5 nerve root compression. METHODS: Protein gene product 9.5 (PGP9.5) was used as a neuronal marker. Rat L5 nerve roots were compressed and then given Yiqi Huayu Decoction, respectively, 10, 20, and 30 days after surgery. The soleus muscles were harvested on day 60 and the endothelium regeneration of neuromuscular junctions was observed by immunohistochemistry and confocal laser scanning microscopy. Α-bungarotoxin (α-BTX) fluorescent binding agent showed motor endplates, and NIH image analysis technique was used to determine the overlapping area of peripheral nerve and motor endplate. Results: After the denervation of the muscle, the peripheral nerves of the Yiqihuayufang group accumulated at the neuromuscular junction, and the sprouting and extension were significantly earlier than those of the control group. During the period of nerve regeneration and repair, the speed and extent of overlap of peripheral nerves and motor end plates and the reconstruction of neuromuscular junctions were also significantly better in the Yiqi Huayu Recipe group than in the control group. Conclusion: Yiqi Huayu Recipe can promote the proliferation of neurons and enhance their regeneration, accelerate the reconstruction of neuromuscular junctions and significantly shorten the process of nerve regeneration.
