目的探讨一种南极细菌胞外多糖体外抗单纯疱疹病毒Ⅰ型(HSV-Ⅰ)活性。方法采用CPE观察与MTT比色法测定南极细菌胞外多糖对人角膜上皮细胞(HCEC)的毒性作用和抗HSV-Ⅰ作用,计算细胞存活率、病毒抑制率及病毒滴度。结果南极细菌胞外多糖对HCEC细胞毒性小,在低浓度下明显促进细胞生长,12.5μg·mL~(-1)浓度下存活细胞量是细胞对照组的146.88%;对HSV-Ⅰ有明显的抑制作用,实验最佳病毒抑制浓度为200μg·mL~(-1)。结论此南极细菌胞外多糖具有显著的低毒、促进细胞生长和抗HSV-Ⅰ活性。
Objective To investigate the anti-herpes simplex virus type 1 (HSV-Ⅰ) activity of Antarctic bacteria exopolysaccharide in vitro. Methods CPE and MTT assay were used to determine the cytotoxicity and anti-HSV-Ⅰ activity of extracellular polysaccharide of human Antarctic bacteria on human corneal epithelial cells (HCECs). The cell viability, virus inhibitory rate and virus titer were calculated. Results Antarctic bacterial extracellular polysaccharides were less toxic to HCEC cells and significantly promoted the cell growth at low concentration. The number of viable cells at 12.5 μg · mL -1 was 146.88% of the cell control group, and HSV-Ⅰ was significantly Inhibition, the best experimental virus inhibition concentration of 200μg · mL -1. Conclusion This Antarctic bacterial extracellular polysaccharide has significant low toxicity, promote cell growth and anti-HSV-Ⅰ activity.
