目的:建立反相高效液相色谱法测定血浆中阿替洛尔浓度。方法:以乙酸乙酯-异丙醇(8:1)为提取溶剂,液-液萃取法处理血浆样品。色谱柱:Phenomenex C18;流动相:0.05 mol·L-1磷酸二氢钠溶液(pH 3.2)-乙腈(90:10)。检测波长:λex =230 nm,λem=310 nm。流速:1.0 ml·min-1。结果:线性范围4.6-883.2 ng·ml-1;最低检测浓度为2.3 ng·ml-1;绝对回收率为95.1%;日内RSD小于3.9%,日间RSD小于5.5%。结论:方法灵敏准确,简便可行,适于阿替洛尔的治疗药物浓度监测。
Objective: To establish a reversed-phase high performance liquid chromatographic method for the determination of atenolol in plasma. Methods: Plasma samples were extracted by liquid - liquid extraction with ethyl acetate - isopropanol (8: 1) as extraction solvent. Column: Phenomenex C18; mobile phase: 0.05 mol·L-1 sodium dihydrogen phosphate solution (pH 3.2) -acetonitrile (90:10). Detection wavelength: λex = 230 nm, λem = 310 nm. Flow rate: 1.0 ml · min-1. Results: The linear range was 4.6-883.2 ng · ml-1. The minimum detectable concentration was 2.3 ng · ml-1. The absolute recovery was 95.1%. The intra-day RSD was less than 3.9%. The intraday RSD Less than 5.5%. Conclusion: The method is sensitive and accurate, simple and feasible, suitable for the concentration monitoring of atenolol in therapeutic drugs.
