目的观察洞庭湖日本血吸虫病流行区居民(实验组)和非疫区居民(对照组)的外周血单核细胞(RBMC)在体外培养条件下,用日本血吸虫成虫可溶性抗原(AWA)和5种重组基因疫苗候选抗原刺激增生时细胞因子的产生情况,并评价这些细胞因子的产生与人体对日本血吸虫感染抗性之间的关系。方法将来自洞庭湖日本血吸虫病洲岛型流行区2个岛(青山和严家山)的67名实验组对象,按其暴露于疫水的程度和血吸虫病粪检结果,分为抗性组(有较高的暴露程度但血吸虫粪卵阴性)、易感组(血吸虫粪卵阳性)和不确定组(有较低程度的暴露且血吸虫粪卵阴性),来自非疫区的11名居民作为对照组。将试验组和对照组对象的外周血单核细胞在血吸虫成虫可溶性抗原和5种重组抗原(副肌球蛋白、28 kDa谷胱甘肽-S-转移酶、脂肪酸结合蛋白、22 kDa表皮膜相关蛋白和 3-磷酸甘油醛脱氢酶)的刺激下培养 5 d,用 ELISA法检测培养上清液中的白细胞介素 5(IL-5),白细胞介素10(IL-10)和γ-干扰素(IFN-γ)的含量。结果试验组对象的PBMC在AWA刺激下产生的 3种细胞因子的水平均高于对照组;但在重组抗原刺激下,只有 IL-10的产生高于对照组;抗性组对象的 PBMC?
OBJECTIVE To observe the peripheral blood mononuclear cells (RBMC) of residents (experimental group) and non-epidemic residents (control group) of schistosomiasis endemic areas of Schistosoma japonicum in Dongting Lake in vitro culture conditions with adult Schistosoma japonicum soluble antigens (AWA) Recombinant gene vaccine candidate antigens stimulate the proliferation of cytokines, and evaluate the relationship between the production of these cytokines and human infection of Schistosoma japonicum. Methods Sixty-seven experimental subjects from the two islands of Schistosoma japonicum endemic island of Schistosoma japonicum in Dongting Lake were divided into three groups according to their degree of exposure to water and schistosomiasis test, Had higher exposure but negative for schistosome fecal egg), susceptible group (positive for schistosome fecal egg), and uncertain group (exposed to a lesser extent and negative for schistosome fecal egg), and 11 residents from the pest free area served as controls group. Peripheral blood mononuclear cells of the experimental and control subjects were associated with soluble antigen of adult schistosomes and five recombinant antigens (paramyosin, 28 kDa glutathione-S-transferase, fatty acid binding protein, 22 kDa epidermal membrane Protein and glyceraldehyde-3-phosphate dehydrogenase) for 5 days. IL-5, IL-10 and γ- Interferon (IFN-γ) content. Results The levels of three cytokines produced by AWA stimulated PBMCs of experimental group were higher than that of control group. However, only the production of IL-10 was higher than that of control group stimulated by recombinant antigen.
