目的研究刺老苞根皮黄酮类化合物对维甲酸致大鼠骨质疏松的影响。方法将60只大鼠随机分成对照组,模型组,刺老苞根皮黄酮类化合物高、中、低剂量(10、20、30 mg/kg)组和仙灵骨葆胶囊(20 mg/kg)阳性对照组,每组10只。各组以ig维甲酸70 mg/kg造模后,各给药组分别ig给予相应药物,每日1次,连续8周。取血清检测相关生化指标,进行骨组织切片检查和骨生化指标检测,测定股骨长度、宽度及质量。结果与对照组比较,模型组大鼠股骨质量、尺骨羟脯氨酸(Hyp)和骨钙的量明显降低,胫骨骨髓腔中脂肪组织增多(P<0.01),血浆胆固醇(TC)水平升高、碱性磷酸酶(ALP)和高密度脂蛋白-胆固醇(HDL-C)水平降低(P<0.01)。与模型组比较,刺老苞根皮黄酮类化合物各剂量组能有效增加骨质量和提高骨质的量(P<0.01),减少骨髓腔中脂肪的量,升高ALP和HDL-C的量(P<0.01)。结论刺老苞根皮黄酮类化合物可有效对抗维甲酸引起的大鼠骨生长抑制及骨丢失。
Objective To study the effects of Radix Gentianae flavonoids on rat osteoporosis caused by retinoic acid. Methods Sixty rats were randomly divided into control group, model group, high, medium and low doses (10,20 and 30 mg / kg) of epidermis flavonoids, and Xianling Gubao Capsule (20 mg / kg ) Positive control group, 10 in each group. Each group with ig retinoic acid 70 mg / kg modeling, the respective drug group were given ig corresponding drugs, once daily for 8 weeks. Serum detection of biochemical indicators, bone tissue biopsy and bone biochemical detection, determination of femur length, width and quality. Results Compared with the control group, the femoral mass, hydroxyproline (Hyp) and calcium in the model group were significantly decreased, the adipose tissue in the tibia medullary cavity increased (P <0.01) and the level of plasma cholesterol (TC) , Alkaline phosphatase (ALP) and high density lipoprotein-cholesterol (HDL-C) decreased (P <0.01). Compared with the model group, each dose of Eupatorium officinale flavones could effectively increase the bone mass and increase the amount of bone mass (P <0.01), decrease the amount of fat in the bone marrow cavity and increase the amount of ALP and HDL-C (P <0.01). Conclusion The root epidermal flavonoids of Eupatorium brasiliensis can effectively inhibit the growth inhibition and bone loss induced by retinoic acid in rats.
