目的:为当归褐斑病菌(Septoria sp.)的深入研究和防治提供理论依据。方法:在当归生长季节系统观察和描述症状发展变化过程。用平皿培养法和悬滴法测定温度、光照、营养和pH值对菌丝生长、分生孢子萌发和产孢的影响,用硫酸法控制小容器相对湿度测定湿度对孢子萌发的影响。结果和结论:该菌丝生长、分生孢子萌发和产孢的温限分别为5~30℃(最适15~25℃)、5~30℃(最适20℃)和5~25℃(最适15℃);连续光照有利于此菌的生长、萌发和产孢;此菌在75%以上相对湿度中均可萌发,以水中萌发最好;菌丝生长期需pH 4.0~10.0,孢子萌发pH 4.51~9.19,两者最适pH均为5.5,产孢pH 4.5~7.5,以pH 6.0产孢量最大;当归叶片浸渍液和葡萄糖液对孢子萌发有较强的促进作用,而蔗糖液和土壤浸渍液则有抑制作用。该菌菌丝生长和孢子萌发速度均缓慢,因此该病害在气温偏低及持续阴雨结露条件下发生较重。
Objective: To provide a theoretical basis for the further study and prevention of Septoria sp. Methods: During the growth season of Angelica Sinensis, observe and describe the process of symptom development and change. The effects of temperature, light, nutrition and pH value on mycelium growth, conidial germination and sporulation were determined by plating culture and hanging drop method. The relative humidity of small container was determined by sulfuric acid method to determine the effect of humidity on spore germination. RESULTS AND CONCLUSION: The temperature limits of mycelial growth, conidial germination and sporulation were 5 ~ 30 ℃ (optimum 15 ~ 25 ℃), 5 ~ 30 ℃ (optimum 20 ℃) and 5 ~ 25 ℃ The optimum temperature is 15 ℃). The continuous illumination is good for the growth, germination and sporulation of this strain. The strain can germinate at 75% RH or more and germinate best in water. The optimum pH value was 5.5 and the sporulation pH was 4.5-7.5. The spore germination rate was the highest at pH 6.0. Angelica leaves immersion solution and glucose solution had a strong promoting effect on the spore germination, while the sucrose solution And soil immersion liquid is inhibited. The mycelium growth and spore germination rate are slow, so the disease in the temperature is low and continued heavy rain condensation occurred.
