[目的]研究曲克芦丁(Troxerutin)与人血清白蛋白(HSA)的相互结合作用。[方法]以HSA为荧光剂,Trox-erutin为荧光猝灭剂,在激发波长为220 nm,发射波长为345 nm下测定两者结合后的荧光强度,推测其猝灭机理;分别测定相同浓度的HSA的荧光发射光谱图和的紫外可见吸收光谱图,来推断两者结合的作用力类型。[结果]Troxerutin与HSA的相互作用为静态猝灭过程,平衡常数K0(25℃)=6.18×105L/mol、K0(37℃)=3.76×105L/mol,供体(HSA)与受体(Troxerutin)间的作用距离r=4.66 nm。Troxerutin和HSA的作用力类型为疏水作用力。[结论]通过两种分析方法清楚的研究了曲克芦丁与血清白蛋白的相互作用。
[Objective] To study the interaction between Troxerutin and human serum albumin (HSA). [Method] With HSA as fluorescent agent and Trox-erutin as fluorescence quencher, the fluorescence intensity of both of them was measured at an excitation wavelength of 220 nm and an emission wavelength of 345 nm, and its quenching mechanism was presumed. The same concentration Of the HSA fluorescence emission spectra and UV-Vis absorption spectra to infer the type of force the combination of the two. [Result] The interaction between Troxerutin and HSA was static quenching. The equilibrium constants K0 (25 ℃) = 6.18 × 105 L / mol, K0 (37 ℃) = 3.76 × 105 L / mol, Troxerutin) the distance between the r = 4.66 nm. The force type of Troxerutin and HSA is hydrophobic. [Conclusion] The interaction between troxerutin and serum albumin was clearly studied by two analytical methods.
