目的建立兔肝硬化动物实验模型,在此基础上建立恶性肿瘤肝转移模型,探讨肝硬化对恶性肿瘤肝转移过程中的影响及其机制。方法采用CCL4皮下注射法建立兔肝硬化模型,随机分为实验组和对照组成,实验组用CCL4溶液皮下注射,对照组给予等量橄榄油。每3周采集兔血和小块肝组织,进行生化指标检测和肝脏HE染色病理学观察。明确肝硬化形成后,分别通过经胃壁、脾脏接种VX2肿瘤细胞,观察肝硬化兔与正常肝脏兔接种VX2肿瘤细胞后肝转移发生率及肿瘤生长情况。结果 (1)实验9周末HE染色可见肝硬化典型病理变化假小叶形成,肝硬化模型建立成功。(2)肝硬化组恶性肿瘤肝转移率明显低于对照组(P<0.05)。结论 CCL4溶液皮下注射成功诱导建立了兔肝硬化模型,并在此基础上建立了恶性肿瘤肝转移模型,实验结果提示肝硬化对恶性肿瘤的肝转移具有一定的抑制作用。
Objective To establish animal model of liver cirrhosis in rabbits and to establish a model of liver metastasis of malignant tumor on the basis of this study and to explore the influence of cirrhosis on liver metastasis of malignant tumor and its mechanism. Methods CCL4 subcutaneous injection was used to establish the model of rabbit cirrhosis. The model group was randomly divided into experimental group and control group. The experimental group was subcutaneously injected with CCL4 solution and the control group was given the same amount of olive oil. Rabbit blood and small pieces of liver tissue were collected every 3 weeks for biochemical detection and pathological observation of liver HE staining. VX2 tumor cells were inoculated through the stomach wall and spleen respectively after the formation of cirrhosis was confirmed. The incidence of liver metastasis and tumor growth after VX2 tumor cells were inoculated in cirrhosis rabbits and normal liver rabbits were observed. Results (1) At the end of the 9th week of experiment, the typical pathological changes of pseudolobules were observed by HE staining. The model of liver cirrhosis was established successfully. (2) The liver metastasis rate of malignant tumor in cirrhosis group was significantly lower than that in control group (P <0.05). Conclusion The CCL4 solution induced by subcutaneous injection successfully established rabbit model of liver cirrhosis and established a model of liver metastasis of malignant tumor. The experimental results suggest that cirrhosis of the liver has a certain inhibitory effect on the liver metastasis of malignant tumor.
