目的观察干细胞因子(stem cell factor,SCF)对高糖环境下豚鼠膀胱Cajal样间质细胞c-kit mRNA及蛋白的表达影响。方法胶原酶消化法原代分离培养豚鼠膀胱Cajal样间质细胞,培养24 h后分别加入葡萄糖浓度为5 mmol/L(正常对照组)、15 mmol/L(高糖组)培养基培养72 h后,高糖组分为高糖对照组、SCF干预组(50 ng/ml、100 ng/ml),分别继续培养24 h、72 h,应用RT-PCR、激光扫描共聚焦显微镜(LSCM)分别检测膀胱Cajal样间质细胞c-kit mRNA和蛋白表达变化。结果高糖对照组较正常对照组c-kit mRNA及蛋白表达均明显下降。24 h组中,SCF50 ng/ml较高糖对照组c-kit mRNA(P>0.05)和蛋白表达(P>0.05)差异不大,SCF100 ng/ml较高糖对照组c-kit mRNA(P<0.05)和蛋白表达(P<0.05)升高;72 h组中,SCF50 ng/ml组、SCF100 ng/ml组较高糖对照组c-kit mRNA(P<0.01)和蛋白表达均明显升高(P<0.01)。结论高浓度SCF可促进已发生形态、机能变化的离体膀胱Cajal样间质细胞c-kit表达上调。
