[目的]探讨鸡血藤血清对人肺癌PG细胞株的增殖抑制及周期阻滞作用。[方法]将肺癌PG细胞分为空白组、鸡血藤组、鸡血藤+顺铂组、顺铂组,四唑盐比色法(MTT)实验中各组根据血清浓度的不同分为20%、10%、5%、2.5%、4组,观察各组PG细胞的增殖抑制作用;HE染色法、免疫荧光法和流式细胞术实验中各组血清浓度为20%,采用光镜和激光共聚焦显微镜观察各组PG细胞形态,流式细胞仪检测细胞周期。[结果]MTT法中5%、10%、20%鸡血藤组细胞的增殖抑制明显,具有时间和剂量依赖性,但低于顺铂组和鸡血藤+顺铂组,鸡血藤+顺铂组PG细胞的增殖抑制率与顺铂组比较,差异无统计学意义(P>0.05)。光镜和激光共聚焦显微镜下鸡血藤组细胞的凋亡程度明显,但低于顺铂组和鸡血藤+顺铂组。与空白组比较,鸡血藤组PG细胞的G_2/M期比例增加(P<0.05);顺铂组和鸡血藤+顺铂组的S和G_2/M期比例增加(P<0.01),G_0/G_1期降低(P<0.01)。顺铂组和鸡血藤+顺铂组较鸡血藤组的S期和G_2/M期比例增加(P<0.01),G_0/G_1期降低(P<0.01)。鸡血藤+顺铂组PG细胞的各期与顺铂组比较,差异无统计学意义(P>0.05)。[结论]鸡血藤血清通过引起细胞周期G_2/M期阻滞,从而抑制细胞增殖,这与顺铂阻滞细胞于G_2/M和S期不同。
[Objective] To investigate the inhibitory effect of Antheraea sera on proliferation and cell cycle arrest of human lung cancer PG cell line. [Methods] The lung cancer PG cells were divided into blank group, Millettia group, Millettia + cisplatin group, cisplatin group and tetrazolium salt colorimetry (MTT). According to the serum concentration, The proliferation of PG cells in each group was observed by HE staining, immunofluorescence and flow cytometry. The serum concentration of each group was 20% The morphology of PG cells in each group was observed by confocal laser scanning microscope, and the cell cycle was detected by flow cytometry. [Result] The MTT assay showed that the proliferation of 5%, 10%, 20% Jixueteng group was significantly inhibited in a time and dose dependent manner, but lower than that in the Cisplatin group, Millettia plus Cisplatin group, The inhibitory rate of proliferation of PG cells in cisplatin group was not significantly different from that in cisplatin group (P> 0.05). Light microscope and confocal laser scanning microscope spatholobus cells apoptosis significantly, but lower than the cisplatin group and Millettia + cisplatin group. Compared with the blank group, the proportion of G 2 / M phase in PG cells increased (P <0.05), and the proportion of S and G 2 / M phases increased in cisplatin-treated and Millettia-treated groups (P <0.01) G_0 / G_1 phase decreased (P <0.01). The ratio of S phase and G 2 / M phase increased in Cisplatin group and Millettia group compared with that in Millettia group (P <0.01), but decreased in G0 / G1 phase group (P <0.01). Millettia and cisplatin group of PG cells compared with the cisplatin group, the difference was not statistically significant (P> 0.05). [Conclusion] The spatholobus serovars could inhibit the cell proliferation by arresting the G 2 / M phase of the cell cycle, which was different from that of cisplatin blocking cells in G 2 / M and S phase.
