利用噬菌体展示技术,发现wssv021是与病毒基因组同源重复区中一个包含高度保守结构域的小重复片断DNA结合的蛋白质的编码基因,可能参与病毒复制或调控.时相转录分析发现,wssv021属于病毒感染的早期基因.将wssv021基因克隆到pQE-30表达载体,在E.coliXL1-Blue中进行融合表达.凝胶阻滞分析显示,体外表达的(His)6-WSSV021蛋白可以和同源重复区中小重复片断DNA特异性相互作用.
Using phage display technology, wssv021was found to be a gene encoding a protein that binds to a highly conserved small repeat-containing DNA in the homologous repeat region of the viral genome, possibly involved in virus replication or regulation.Western blot analysis revealed that wssv021is a virus Infected early gene.The wssv021 gene was cloned into pQE-30 expression vector and expressed in E.coli XL1-Blue.The results of gel blocking analysis showed that the (His) 6-WSSV021 protein expressed in vitro could hybridize with the homologous repeat region Small and medium repeat DNA-specific interactions.
