目的 探讨抑制宫颈癌中中性粒细胞相关载脂蛋白(NGAL)基因的表达对癌细胞增殖及凋亡的影响及机制.方法 RT-PCR检测NGAL基因在宫颈癌组织中的mRNA表达;人宫颈癌HeLa细胞分为空白组、阴性对照组和NGAL转染组,转染48 h后,Western bloting检测各组细胞中NGAL、活化的含半胱氨酸的天冬氨酸蛋白水解酶(Cleaved caspase3)、B细胞淋巴瘤/白血病-2(Bcl-2)、β-连环蛋白(β-catenin)、细胞周期素D1(Cyclin D1)蛋白表达;CCK8实验和流式细胞仪分别检测细胞的增殖和凋亡情况.结果 宫颈癌组织中NGAL的mRNA表达显著高于癌旁组织(P<0.01);NGAL转染组NGAL的蛋白表达显著降低;NGAL转染组细胞存活率及Bcl-2、β-catenin、Cyclin D1蛋白表达均显著低于空白组(P<0.01);细胞凋亡率及Cleaved caspase3蛋白表达显著高于空白组(P<0.01).结论 抑制宫颈癌中NGAL基因的表达可显著降低癌细胞的增殖及诱导细胞凋亡,其机制与下调Wnt/β-catenin信号通路有关.“,”Objective To investigate the effect and mechanism of NGAL gene expression inhibited on proliferation and apoptosis of cervical cancer.Methods The mRNA expression of NGAL gene was detected in cervical carcinoma by RT-PCR.The human cervical carcinoma HeLa cells were divided into control group,negative control group and NGAL transfection group.NGAL,Cleaved Caspase3,Bcl-2,β-catenin,Cyclin D1 protein expression transfected for 48h were detected by Western bloting.CCK8 assay and flow cytometry were used to detect cell proliferation and apoptosis.Results The mRNA expression of NGAL in cervical carcinoma was significantly higher than paracancerous tissue (P<0.01).The expression of NGAL protein in NGAL transfected group decreased significantly,cell survival rate and the expression of Bcl-2,β-catenin,Cyclin D1 protein in NGAL transfection group were significantly lower than control group(P<0.01).The cell apoptosis rate and the expression of Cleaved Caspase3 protein were significantly higher than the control group (P<0.01).Conclusion The expression of NGAL gene inhibited in cervical cancer could significantly reduce the proliferation and induce apoptosis of cancer cells,and the mechanism is related to decrease regulation of Wnt/β-catenin signaling pathway.
