目的观察肾病Ⅰ号方总皂苷对单侧输尿管结扎(UUO)大鼠肾纤维化的影响,阐明其抗肾纤维化的作用机制,探索中药复方药效物质基础的研究思路。方法建立UUO大鼠模型,30只大鼠随机分为5组,每组6只:正常组、模型组、贝那普利组、总皂苷组、肾病Ⅰ号方组。于术后两周随机处死大鼠,取左侧肾组织。观察干预后各组大鼠一般情况变化,测定24小时尿蛋白定量、血清肌酐(Scr)、血清尿素氮(BUN)的含量,光镜下观察肾组织纤维化的病理改变,免疫组织化学法检测肾组织胶原Ⅳ(COL-IV)的表达情况。结果肾病Ⅰ号方总皂苷能显著降低Scr、BUN,减轻尿蛋白,减少细胞外基质COL-IV的阳性表达(P<0.05),改善肾小球硬化程度,与贝那普利组和肾病Ⅰ号方组比较无明显差异(P>0.05)。结论肾病Ⅰ号方总皂苷可以抑制细胞外基质COL-IV的合成,减少细胞外基质的积聚,从而达到抗肾纤维化的作用。
Objective To observe the effect of total saponin of nephrosis No.1 on renal fibrosis in rats with unilateral ureteral obstruction (UUO), clarify its mechanism of anti-renal fibrosis and explore the research ideas of the pharmacodynamic substance of traditional Chinese medicine. Methods UUO rat model was established. Thirty rats were randomly divided into five groups with 6 rats in each group: normal group, model group, benazepril group, total saponin group and nephrotic group Ⅰ. Two weeks after surgery, rats were sacrificed randomly and the left renal tissue was obtained. The general changes of rats in each group were observed after intervention. The content of urinary protein, serum creatinine (Scr) and serum urea nitrogen (BUN) in 24 hours were measured. The pathological changes of renal fibrosis were observed under light microscope. Immunohistochemistry Renal collagen Ⅳ (COL-IV) expression. Results Total Saponin Ⅰ could significantly reduce Scr, BUN, reduce urinary protein, decrease the expression of extracellular matrix COL-IV (P <0.05), and improve the degree of glomerulosclerosis, and benazepril group and nephropathy Ⅰ No. square group no significant difference (P> 0.05). Conclusion Nephropathy Ⅰ total saponin can inhibit extracellular matrix COL-IV synthesis, reduce the accumulation of extracellular matrix, so as to achieve anti-renal fibrosis.
