组织切片染色中组织和染料的吸引方式可分为五种类型。其中之一是染料和组织均电离,染色反应基本上属于酸碱结合反应,带阳电荷的组织与带阴电荷的染料结合;带阴电荷的组织则与带阳电荷的染料结合。细胞核带阴电荷,它与带阳电荷的染料(碱性染料)结合,而细胞浆中的蛋白质或胶元纤维带阳电荷,与带阴电荷的染料(酸性染料)相结合。
