穿山龙抗急性痛风性关节炎的肾脏代谢组学研究

目的:通过肾脏代谢组学研究穿山龙总皂苷对急性痛风性关节炎模型大鼠的防治作用,并找到其相关的潜在生物标志物和代谢通路。方法:采用尿酸钠诱导的急性痛风性关节炎模型并运用超高液相色谱-飞行时间-质谱(UPLC-TOF-MS)结合模式识别方法探讨穿山龙总皂苷干预急性痛风性关节炎的潜在生物标志物及其相关代谢通路。结果:鉴别出了7个共同的潜在生物标志物,其中穿山龙对正常大鼠的潜在干预靶点中,4个上调,3个下调;穿山龙对急性痛风性关节炎模型大鼠的潜在干预靶点中,5个代谢物在尿酸钠诱导下被上调,2个代谢物被下调,而穿山龙总皂苷表现出纠正软骨素、甘油磷酰胆碱、乙二酸、廿二碳六烯酸、牛磺酸这5个代谢物异常表达的趋势。相关性最强的代谢通路是牛磺酸和亚牛磺酸代谢、乙醛酸和二羧酸代谢,其涉及的潜在内源性生物标志物分别是牛磺酸和乙二酸。结论:穿山龙主要通过干预急性痛风性关节炎状态下和生理状态下肾脏中牛磺酸和乙二酸的变化,以及所参与的牛磺酸和亚牛磺酸代谢、乙醛酸和二羧酸代谢,实现防治急性痛风性关节炎作用。 OBJECTIVE: To study the prevention and treatment of total gypenosides in rats with acute gouty arthritis by renal metabonomics and to find out its potential biomarkers and metabolic pathways. Methods: UA-induced acute gouty arthritis model was established and the potential biomarkers for the intervention of Asiasatis Saponins in acute gouty arthritis were investigated by UPLC-TOF-MS combined with pattern recognition Matter and its related metabolic pathways. Results: Seven potential biomarkers were identified. Among them, four of them were up-regulated and three of them were potential targets for intervention in normal rats. Potential intervention targets of S. auricula in acute gouty arthritis rats , Five metabolites were up-regulated under uric acid sodium induction and two metabolites were down-regulated while S. aureus showed signs of correct chondroitin, glycerophosphorylcholine, oxalic acid, docosahexaenoic acid, taurine The five metabolites of acid were abnormally expressed. The most relevant metabolic pathways are the metabolism of taurine and hypotaurine, the metabolism of glyoxylate and dicarboxylic acid, and the underlying endogenous biomarkers involved are taurine and oxalic acid, respectively. CONCLUSIONS: It is mainly through the intervention of rheumatoid arthritis in the state and the physiological state of taurine and oxalic acid in the kidney changes, as well as the participation of taurine and hypotaurine metabolism, glyoxylate and dicarboxylic acid Metabolism, to achieve the effect of prevention and treatment of acute gouty arthritis.
