目的:检测表皮生长因子受体Ⅲ型突变体(EGFRvⅢ)在食管癌组织中的表达,探讨其与食管癌发生、发展的关系。方法:采用免疫组化和蛋白质印迹实验检测EGFRvⅢ在66例人食管癌组织的表达情况,评价EGFRvⅢ表达在食管癌患者的性别、年龄分布、肿瘤大小、生长方式、TNM分期和淋巴结转移等临床病理参数的表达分布情况。结果:免疫组化结果显示EGFRvⅢ主要分布于胞膜及胞质内,在肿瘤及正常组织表达的平均净灰度值分别为22.46±4.21和5.54±3.01,两者相比差异有统计学意义,t=6.701,P=0.001。蛋白质印迹实验结果显示,EGFRvⅢ在肿瘤及瘤旁正常组织中表达的平均净灰度值分别为0.828±0.15和0.083±0.049,两者相比差异有统计学意义,t=9.362,P=0.000。两种检验方法均显示,不同性别、年龄、肿瘤大小、生长方式组间EGFRvⅢ表达,差异无统计学意义(P>0.05),TNM分期、淋巴结转移和病理分级中EGFRvⅢ表达差异均有统计学意义,P<0.05。EGFRvⅢ在两种实验方法的一致性检验(r=0.917,P<0.001),提示两种方法结果一致性好。结论:EGFRvⅢ可能参与食管癌发生,并在食管癌的侵袭和转移中起重要作用。
Objective: To detect the expression of epidermal growth factor receptor type Ⅲ mutant (EGFRv Ⅲ) in esophageal carcinoma and to explore its relationship with the occurrence and development of esophageal cancer. Methods: Immunohistochemistry and Western blotting were used to detect the expression of EGFRv Ⅲ in 66 human esophageal carcinomas and to evaluate the clinical and pathological features of EGFRv Ⅲ in esophageal cancer patients, including gender, age distribution, tumor size, growth pattern, TNM stage and lymph node metastasis Parameters of the distribution of the situation. Results: The results of immunohistochemistry showed that the average net gray value of EGFRv Ⅲ in the membrane and cytoplasm were 22.46 ± 4.21 and 5.54 ± 3.01, respectively. There was significant difference between the two groups t = 6.701, P = 0.001. The results of Western blotting showed that the average net gray value of EGFRv Ⅲ expressed in tumor and adjacent normal tissues were 0.828 ± 0.15 and 0.083 ± 0.049, respectively, with statistical significance (t = 9.362, P = 0.000). There was no significant difference in the expression of EGFRvIII between the two groups according to gender, age, tumor size and growth pattern (P> 0.05). There were significant differences in the expression of EGFRvIII in TNM stage, lymph node metastasis and pathological grading , P <0.05. EGFRv Ⅲ consistency test in the two experimental methods (r = 0.917, P <0.001), suggesting that the two methods have good agreement. Conclusion: EGFRv Ⅲ may be involved in esophageal carcinogenesis and play an important role in the invasion and metastasis of esophageal cancer.
