目的研究藏花素诱导膀胱移行细胞癌T24细胞凋亡的作用及分子机制。方法通过MTT比色法分析藏花素对T24细胞增殖的影响;应用光镜观察细胞形态学的改变;Annexin V-FITC/PI荧光双标流式细胞术检测T24细胞周期动力学和凋亡诱导率的影响,RT-PCR和Western blot检测凋亡相关基因Bcl-2、Bax的表达变化。结果藏花素能明显抑制T24细胞的增殖,并可导致其形态学的改变;随藏花素作用时间的延长,G0/G1期细胞比例逐渐增加,S期和G2/M期细胞比例逐渐减少,早期凋亡细胞逐渐增多,同时可以上调Bax和下调Bcl-2的基因表达水平。结论藏花素可明显抑制T24细胞的生长,诱导肿瘤细胞凋亡,可能与Bcl-2和Bax等凋亡调控基因有关。
Objective To study the effect and molecular mechanism of cyanidin on apoptosis of bladder transitional cell carcinoma T24 cells. Methods The effect of crocin on proliferation of T24 cells was analyzed by MTT colorimetry; the changes of cell morphology were observed by light microscope; the cell cycle kinetics and apoptosis induction of T24 were detected by Annexin V-FITC/PI double-label flow cytometry. The effect of rate was detected by RT-PCR and Western blot to detect the expression of apoptosis-related genes Bcl-2 and Bax. RESULTS: Chlorophyllin can significantly inhibit the proliferation of T24 cells and lead to its morphological changes. With the prolongation of florescence time, the proportion of cells in G0/G1 phase gradually increased, and the proportion of cells in S phase and G2/M phase gradually decreased. The number of early apoptotic cells gradually increased, while it could up-regulate Bax and down-regulate Bcl-2 gene expression. Conclusion Chlorophyllin can obviously inhibit the growth of T24 cells and induce the apoptosis of tumor cells, which may be related to the apoptosis regulating genes such as Bcl-2 and Bax.
