目的研究二苯甲酮-3(benzophenone-3,BP-3)对绒毛外滋养层(HTR-8/Svneo)细胞的H19及mi R-675 m RNA表达的影响,并探讨非编码RNA H19在BP-3抑制HTR-8/Svneo细胞增殖中的作用。方法将处于对数生长期的HTR-8/Svneo细胞分别暴露于含终浓度为0(溶剂对照)、0.01、0.1、1 mg/L BP-3的培养基或转染试剂(脂质体2000和mi R-675抑制剂或H19抑制剂)中培养24 h。采用Real-time PCR检测H19、mi R-675 m RNA的表达水平;CCK-8法检测HTR-8/Svneo细胞的存活率。结果与溶剂对照组相比,1 mg/L BP-3染毒组HTR-8/Svneo后H19及mi R-675的表达水平均降低,差异均有统计学意义(P<0.01);且随着BP-3染毒剂量的升高,HTR-8/Svneo细胞的H19及mi R-675的表达水平均呈下降趋势。与抑制剂对照组相比,加入H19抑制剂后HTR-8/Svneo细胞的存活率和mi R-675 m RNA的表达水平均降低;加入mi R-675抑制剂后HTR-8/Svneo细胞的存活率均降低,差异有统计学意义(P<0.01)。结论 H19通过mi R-675在BP-3抑制HTR-8/Svneo细胞增殖中的发挥作用。
Objective To investigate the effect of benzophenone-3 (BP-3) on the expression of H19 and mi R-675 m RNA in HTR-8 / Svneo cells and to explore the role of non-coding RNA H19 BP-3 inhibits the proliferation of HTR-8 / Svneo cells. Conclusion H19 plays a role in the inhibition of HTR-8 / Svneo cell proliferation by mi R-675 by BP-3.
