目的:考察桂枝茯苓胶囊及其15个活性成分组合物对大鼠盆腔炎模型的影响,初步探讨其对盆腔炎的治疗作用。方法:90只雌性SD大鼠分为假手术组、模型组,模型组采用大肠埃希菌造成大鼠盆腔炎模型,将造模成功的大鼠随机分为模型组,桂枝茯苓胶囊组(250 mg·kg~(-1)),15个成分组合物低、中、高剂量组(75,150,300 mg·kg~(-1)),每组15只。桂枝茯苓胶囊及其成分组合物ig给药后15 d后将大鼠处死,取子宫后称重并计算子宫质量系数;取血后测血清肿瘤坏死因子-α(TNF-α),白细胞介素-2(IL-2)的含量及血浆免疫球蛋白A(Ig A),Ig G和Ig M的含量;苏木素-伊红(HE)染色检测大鼠子宫组织病理变化。结果:与假手术组比较,模型组大鼠子宫系数明显升高,血浆Ig A,Ig G和Ig M的含量明显降低,子宫病理综合评分明显升高(P<0.01);与模型组比较,桂枝茯苓胶囊及活性成分组合物高、中、低剂量组均能显著降低盆腔炎大鼠的子宫系数,能明显降低盆腔炎模型大鼠血清TNF-α含量,升高IL-2含量,并提高血浆Ig A,Ig G和Ig M的含量,对子宫病理综合评分也有显著性降低(P<0.05,P<0.01)。结论:15个活性成分组合物与桂枝茯苓胶囊均有明显的抗实验性大鼠盆腔炎作用,提示15个活性成分可能是桂枝茯苓胶囊盆腔炎的重要物质基础。
Objective: To investigate the effects of Guizhi Fuling capsule and its 15 active ingredients on pelvic inflammatory disease in rats, and to explore its therapeutic effect on pelvic inflammatory disease. Methods: 90 female SD rats were randomly divided into sham operation group, model group and model group. Pelvic inflammatory disease model was induced by Escherichia coli in rats. The successful rats were randomly divided into model group, Guizhi Fuling capsule group 250 mg · kg ~ (-1)), and 15 low, medium and high dose groups (75, 150 and 300 mg · kg ~ (-1)). The rats were sacrificed 15 days after ig administration of Guizhi Fuling Capsule and its composition. The uterus was weighed and the uterine mass coefficient was calculated. After taking the blood, the levels of serum tumor necrosis factor-α (TNF-α), interleukin The content of IL-2, Ig A, Ig G and Ig M in the uterus were detected by hematoxylin and eosin (HE) staining. Results: Compared with the sham operation group, the uterine coefficient of the model group was significantly increased, the contents of Ig A, Ig G and Ig M in the model group were significantly decreased, and the comprehensive pathological score of the uterus was significantly increased (P <0.01). Compared with the model group, Guizhi Fuling Capsule and active ingredient combination could significantly decrease the uterine coefficient of pelvic inflammatory disease rats, and significantly decrease the content of TNF-α and IL-2 in pelvic inflammatory disease model rats The plasma Ig A, Ig G and Ig M levels were also significantly decreased (P <0.05, P <0.01). CONCLUSION: The 15 active ingredients and Guizhi Fuling Capsule have significant anti-inflammatory effects on experimental rats, suggesting that 15 active ingredients may be the important material basis of Guizhifuling capsule pelvic inflammatory disease.
