目的探讨重组腺病毒介导的转录因子Runx3对PC-3前列腺癌细胞迁移和侵袭的影响。方法以Ad-Runx3感染的PC-3细胞为实验组,Ad-EGFP感染的PC-3细胞为对照组,Western blot检测Runx3-flag的表达,间接免疫荧光分析其定位;MTT法观察Runx3过表达对PC-3细胞生长的影响,Transwell迁移实验检测对PC-3细胞迁移的影响,Matrigel侵袭实验检测对PC-3细胞侵袭的影响。结果 Western blot显示Runx3-flag可表达于PC-3细胞中,间接免疫荧光显示其表达定位于细胞核中;腺病毒介导Runx3过表达对PC-细胞体外生长、迁移及侵袭均具有抑制作用。结论重组腺病毒介导的转录因子Runx3可表达于PC-3细胞核中,其过表达对PC-3细胞体外生长、迁移及侵袭均具有抑制作用,为前列腺癌的治疗提供了新策略。
Objective To investigate the effect of recombinant adenovirus-mediated transcription factor Runx3 on the migration and invasion of PC-3 prostate cancer cells. Methods PC-3 cells infected with Ad-Runx3 were used as experimental group, and PC-3 cells infected with Ad-EGFP were used as control. Runx3-flag expression was detected by Western blot and indirect immunofluorescence staining was used to analyze the expression of Runx3. Runx3 overexpression On the growth of PC-3 cells, the effect of Transwell migration assay on the migration of PC-3 cells and the Matrigel invasion assay on the invasion of PC-3 cells. Results Runx3-flag was expressed in PC-3 cells by Western blot. Indirect immunofluorescence showed that Runx3-flag localized in the nucleus. Adenovirus-mediated overexpression of Runx3 could inhibit the growth, migration and invasion of PC-3 cells in vitro. Conclusion The recombinant adenovirus-mediated transcription factor Runx3 can be expressed in the nucleus of PC-3 cells. The overexpression of Runx3 can inhibit the growth, migration and invasion of PC-3 cells in vitro and provide a new strategy for the treatment of prostate cancer.
