目的:建立山慈菇多糖的含量测定方法。方法:采用苯酚-浓硫酸方法显色,利用紫外分光光度计在488.5nm处进行测定。结果:得到回归方程为:Y=0.4538x-0.0067,r=0.9992,测得山慈菇多糖含量为:14.48%,加样回收率平均为97.8%,RSD值为2.24%(n=9)。结论:用苯酚-浓硫酸法显色,方法简单,可靠,精密度、稳定性及重复性好,可用于山慈菇多糖的含量测定。
Objective: To establish a method for determination of polysaccharides in Pleurotus. Methods: Phenol - concentrated sulfuric acid method was used for colorimetric determination with UV spectrophotometer at 488.5nm. Results: The regression equation was obtained as follows: Y = 0.4538x-0.0067, r = 0.9992. The content of polysaccharides in Fructus Sargassum was 14.48%. The average recovery was 97.8% with RSD value of 2.24% (n = 9). Conclusion: The method of phenol - concentrated sulfuric acid method is simple, reliable, precision, stability and reproducibility, and can be used for the determination of polysaccharides from.
