1982年秋,在太安市效长豇豆田采集病株种子,以播种后所得的病菌为毒源,在苋色蔾上经单斑分离纯化,得一病毒分离物CV-sd。CV-sd侵染长豇豆“红嘴燕”品种所引起的症状为浓绿色脉带,花叶及叶片畸形。该分离物的寄主范围较窄,在测定的11个科的供试植物中,主要侵染豆科和蔾科植物。CV-sd的钝化温度为55°—60℃,稀释限点为10~(-2)。体外存活期为一天(室温),易经汁液传染和桃蚜,豆蚜、棉蚜以非持久方式传播,并经种子传播,其种传率为18.9%。电镜下观察,CV-sd质粒呈线条状,长720-740nm,电镜下观察病组织超薄切片,在受侵细胞质内存在大量风轮状内含体。用CV-sd提纯制品免疫家免所得的抗血清的效价为1/8。血清学测定结果表明,CV-sd与黑眼豇豆花叶病毒有密切的血清学关系,并与豇豆蚜传花叶病毒和菜豆普通花叶病毒有不同程度的血清学关系。据此,作者认为该分离物为黑眼豇豆花叶病毒。
In the autumn of 1982, the diseased cowpea seeds were harvested in the effective cowpea field of Tai’an City. The virus was sown after being sowed. The isolates had a narrower host range. Among the 11 tested plants tested, they mainly infected legumes and Chenopodiaceae plants. Under the electron microscope, the CV-sd plasmid was linear and elongated 720-740nm. The ultrastructure of diseased tissue was observed under electron microscope. There were a large number of wind-like inclusions in the cytoplasm. The titer of the antiserum raised by immunization with CV-SD was about 1/8. The serological results showed that CV-sd had close serological relationship with black-eye cowpea mosaic virus and had different degrees of serological relationship with cowpea aphid-transmitted mosaic virus and common bean mosaic virus. Accordingly, the authors consider the isolate as black-eye cowpea mosaic virus.
