目的:研究人巨细胞病毒(HCMV)感染对人脐静脉内皮细胞(HUVEC)糖基化终产物受体(RAGE)表达的影响,探讨HCMV感染致动脉粥样硬化的作用机制。方法:用HCMV感染HUVEC,采用RT-PCR方法检测不同感染时段细胞RAGEmRNA的表达。结果:HCMV感染HUVEC后,0 h时RAGEmR-NA有基础水平的表达,4 h开始升高,8 h时表达水平明显升高,随感染时间延长,表达量逐渐增高,感染24 h时达高峰,48 h开始回落,72 h表达量仍维持在较高的水平,显著高于0 h。各时段与0 h比较,均有统计学意义(P<0.01)。结论:HCMV感染人脐静脉内皮细胞后能够促进RAGE mRNA的表达,并且呈时间依赖性。HCMV有可能通过上调RAGE介导内皮细胞的炎症反应,促进动脉粥样硬化的发生、发展。
Objective: To investigate the effect of human cytomegalovirus (HCMV) infection on the expression of advanced glycation end products receptor (RAGE) in human umbilical vein endothelial cells (HUVECs) and explore the mechanism of atherosclerosis induced by HCMV infection. Methods: HUVECs were infected with HCMV, and the expression of RAGE mRNA was detected by RT-PCR. Results: The expression of RAGEmR-NA at 0 h after HCMV infection in HUVEC increased from 4 h, and then increased at 8 h. With the prolongation of infection time, the expression of RAGEmR-NA increased gradually and peaked at 24 h 48h began to decline, 72 h expression remained at a high level, significantly higher than 0 h. Compared with 0 h in each time period, there was statistical significance (P <0.01). CONCLUSION: HCMV infect human umbilical vein endothelial cells can promote the expression of RAGE mRNA in a time-dependent manner. HCMV may promote the occurrence and development of atherosclerosis by up-regulating RAGE-mediated endothelial cell inflammation.
