已知大鼠垂体前叶细胞结合促性尕激素释放激素(GnRH)与肌醇磷脂动员有关,后者可产生二酰基甘油和三磷酸肌醇(IP_3)。作者研究了分离的大鼠垂体前叶细胞,胞外给予磷酸肌醇的作用。从鼠龄6~8周雌性SD大鼠分离垂体前叶腺,经抑菌,消化处理后进行细胞培养,3天后进行灌流,并加入各种多磷酸肌醇进行观察。每两分钟采样,用放免法测定LH。并用阳离子交换色谱法测定各种磷酸肌醇,用液问法测定~3H标记的肌醇。结果显示灌流液中加入二磷酸肌醇(IP_2)和IP_3后,可引起LH释放增加。IP_2和IP_3均可使LH的基础分泌量增加,作用呈剂量依赖关系。IP_2和IP_3促分
Rat anterior pituitary cells are known to be associated with mobilization of gnhH and mobilization of inositol, which produces diacylglycerol and inositol triphosphate (IP_3). The authors studied the isolated rat anterior pituitary cells, extracellular phosphoinositide. The anterior pituitary gland of pituitary gland was isolated from 6 to 8 weeks old female Sprague-Dawley rats. After antibacterial and digestion treatment, the cells were cultured and perfused 3 days later, and various inositol polyphosphates were added for observation. Sampling every two minutes, using radioimmunoassay LH. Various phosphoinositides were determined by cation exchange chromatography and ~ 3H labeled myo-inositol was determined by liquid chromatography. The results showed that the addition of diphosphate inositol diphosphate (IP_2) and IP_3 increased the release of LH.
