目的:观察氧化低密度脂蛋白(Oxidative low density lipoprotein,Ox-LDL)对外周血内皮祖细胞(en-dothelial progenitor cells,EPCs)功能的影响,并研究丹参素的保护作用。方法:密度梯度离心法获取外周血单个核细胞,培养7天后,收集贴壁细胞并随机分为对照组,氧化低密度脂蛋白组(100μg.mL-1)及丹参素加氧化低密度脂蛋白组(丹参素浓度分为2、10和50μg.mL-1)。采用流式细胞仪鉴定细胞,然后分别采用MTT比色法、黏附能力测定实验观察EPCs的增殖能力、黏附能力。结果:和对照组相比,Ox-LDL显著损伤了外周血EPCs的黏附能力、增殖能力(P<0.01),经过丹参素干预24h后,显著改善了外周血EPCs的黏附能力(40±6vs50±10,P<0.05)、增殖能力(0.12±0.02vs0.24±0.08,P<0.05)。结论:氧化低密度脂蛋白显著损伤了外周血EPCs的功能,丹参素能保护Ox-LDL损伤后内皮祖细胞的功能。
Objective: To observe the effect of Oxidized low density lipoprotein (Ox-LDL) on the function of peripheral blood endothelial progenitor cells (EPCs) and to study the protective effect of Danshensu. Methods: Peripheral blood mononuclear cells were obtained by density gradient centrifugation. After cultured for 7 days, adherent cells were collected and randomly divided into control group, oxidized low-density lipoprotein group (100 μg·mL-1) and Danshensu plus oxidized low-density lipoprotein. The group (danshensu concentration was divided into 2, 10 and 50 μg·mL-1). The cells were identified by flow cytometry, and the proliferation and adhesion of EPCs were observed by MTT assay and adhesion assay. RESULTS: Compared with the control group, Ox-LDL significantly impaired the adhesion and proliferative capacity of peripheral blood EPCs (P<0.01). After intervention with Danshensu for 24 h, the adherence of peripheral blood EPCs was significantly improved (40±6 vs 50±). 10, P<0.05), proliferation ability (0.12±0.02 vs 0.24±0.08, P<0.05). CONCLUSION: Oxidized low-density lipoprotein significantly impairs the function of peripheral blood EPCs. Danshensu can protect the function of endothelial progenitor cells after Ox-LDL injury.
