目的:比较不同培养条件对白念珠菌芽管形成的影响。方法:分别采用胎牛血清(FCS)、灭活人血清、RPMI1640培养基、DMEM培养基,在不同条件(孵育温度、孵育时长、pH值、起始菌浓度)下进行芽管的诱导培养,并采用结晶紫染色法计算出芽率。结果:相同条件下,FCS比RPMI1640、DMEM培养基诱导出芽率高(P<0.05),而后两者之间无显著性差异(P>0.05);白念珠菌起始菌体浓度为5×10~6 cfu/mL时,于FCS中37℃振荡孵育2h,出芽率及芽管纯度最好;当菌体浓度超过5×10~6 cfu/mL时,随着菌体浓度的增加,出芽率反而下降。结论:优化培养条件可获得较纯的芽管,结晶紫染色法可精确定量检测芽管形成率。
Objective: To compare the effects of different culture conditions on the formation of Candida albicans germ tube. Methods: Induction culture of germ tube was carried out under different conditions (incubation temperature, incubation time, pH value and initial bacterium concentration) with fetal calf serum (FCS), inactivated human serum, RPMI1640 medium and DMEM medium, And the crystal violet staining method to calculate the bud rate. Results: Under the same conditions, FCS had higher germination rate than RPMI1640 and DMEM medium (P <0.05), but there was no significant difference between the two groups (P> 0.05). The initial concentration of Candida albicans was 5 × 10 ~ 6 cfu / mL, incubated in FCS at 37 ℃ for 2h, the budding rate and bud tube purity were the best. When the concentration of bacteria was more than 5 × 10 ~ 6 cfu / mL, the budding rate But declined. Conclusion: The pure bud tube can be obtained by optimizing the culture conditions. The crystal violet staining method can accurately detect the tube formation rate.
