用平衡透析、凝胶色谱等方法研究了牛血清白蛋白(BSA)与PtCl_4~(2-)、顺铂及cis-[Pt(NH_3)_2(H_2O)_2]~(2+)的相互作用。结果表明,顺铂与BSA在25℃作用4小时,未发现BSA结合铂,而在37℃作用20小时后BSA与铂(Ⅱ)有一定的结合。BSA对PtCl_4~(2-)有一个强结合部位,3个弱结合部位,结合常数分别为4.0×10~4及1.1×10~3。BSA对cis-[Pt(NH_3)_2(H_2O)_2]~(2+)有2个专一性结合部位,结合常数K_1=9.0×10~4,有2-3个非专一性结合部位,K_2=2.4×10~2,BSA与cis-[Pt(NH_3)_2(H_2O)_2]~(2+)的反应对BSA是一级反应,速度常数k=0.012分~(-1)。
The interaction between bovine serum albumin (BSA) and PtCl_4 ~ (2-), cisplatin and cis- [Pt (NH_3) _2 (H_2O) _2] 2+ was studied by equilibrium dialysis and gel chromatography. . The results showed that BSA combined with BSA at 25 ℃ for 4 hours showed no binding of platinum to BSA and platinum (Ⅱ) at 37 ℃ for 20 hours. BSA has a strong binding site for PtCl_4 ~ (2-) and three weak binding sites with binding constants of 4.0 × 10 ~ 4 and 1.1 × 10 ~ 3, respectively.
