采用双向电泳(2-DE)技术,对薄皮甜瓜白莎蜜1号和叶色黄化突变体9388-1叶片蛋白质组差异表达进行分析与鉴定。结果表明,在双向电泳图谱上检测到比较清晰的蛋白点共1 136个;蛋白质特异性表达及3倍差异表达的蛋白点分别为89个和102个,其中白莎蜜1号特异性表达的蛋白点有9个,叶色黄化突变体9388-1有80个,二者表达丰度3倍差异的蛋白点各51个;在叶色黄化突变体9388-1中表现上调的蛋白点24个,表现下调的蛋白点27个;从差异表达的蛋白点中筛选19个进行质谱分析和数据库检索,鉴定出参与能量产生与转换蛋白1个、核苷酸运输与代谢蛋白2个、碳水化合物运输与代谢蛋白8个、翻译后修饰及蛋白周转和伴侣蛋白2个、细胞骨架及细胞壁与细胞膜合成蛋白2个,其差异表达均可能与叶色黄化有关。
Two-dimensional electrophoresis (2-DE) technique was used to analyze and identify the differentially expressed proteins in the leaves of the white-skinned melon, Baishayitian 1 and leaf-chlorotic mutant 9388-1. The results showed that a total of 1 136 protein spots were detected clearly in the two-dimensional electrophoresis pattern, and 89 and 102 protein-specificity spots and 3-fold differential spots respectively. Among them, Baisha No. 1 There were 9 protein spots, 80 leaf spot yellowing mutants 9388-1, 51 protein spots with 3-fold difference in abundance, and up-regulated protein spots in yellowing mutant 9388-1 24, down-regulated protein spots 27; screening 19 differentially expressed protein spots for mass spectrometry and database searches, identification of one involved in energy production and translation protein, two nucleotide transport and metabolism proteins, carbohydrates There were 8 transport and metabolism proteins, 2 posttranslational modifications, 2 protein turnover and chaperonins, 2 cytoskeleton and cell wall and membrane protein synthesis, all of which may be related to leaf color yellowing.
