目的 :探讨抗纤复方对体外培养纤维肝大鼠肝细胞基底膜基质生成的影响。方法 :常规培养正常与 DMN纤维肝大鼠肝细胞 ,然后改用抗纤复方原液与抗纤复方药物血清温育肝细胞 48小时后 ,免疫组化 S-P法显色细胞纤维连接蛋白( FN)、层连蛋白 ( LM)和 IV型胶原蛋白 ( IV-C)的变化。结果 :正常肝细胞基底膜基质合成处于抑制状态 ,肝纤维化时肝细胞基底膜基质合成增加。抗纤复方能显著抑制纤维肝肝细胞 F N,L M及 IV-C的表达 ,抗纤复方药物血清效果优于原液。结论 :抗纤复方具有抑制 DMN纤维肝肝细胞基底膜基质的异常合成作用。
Objective: To investigate the effects of Kangxian Fufang on the formation of hepatocyte basal matrix in fibrotic liver in vitro. Methods: Normal liver cells were cultured with normal DMN fibrosis and liver fibrosis was induced by anti-fibrosis compound and anti-fibrosis compound for 48 hours. Immunofluorescence staining was used to detect the expression of fibronectin (FN) Changes in laminin (LM) and type IV collagen (IV-C). Results: The synthesis of basement membrane of normal liver cells was inhibited. The synthesis of basement membrane of liver cells increased during liver fibrosis. Anti-fibrosis compound can significantly inhibit the expression of FN, L M and IV-C in fibrotic liver cells, anti-fibrosis compound serum better than the original solution. Conclusion: Kangxian compound can inhibit the abnormal synthesis of DMN fibroblast basement membrane matrix.
