目的建立测定小鼠血浆神经生长因子(NGF)浓度的同位素示踪沉淀法。方法采用放射性同位素(~(125)I)标记示踪法结合三氯醋酸(TCA)沉淀法测定NGF的浓度。结果本方法的最低检测浓度为2.0 ng/ml,日内、日间变异系数(CV)均小于10%,血浆中NGF在12.5~400 ng/ml浓度范围内均呈良好的相关性,相关系数r=0.9990,血浆中NGF的平均回收率为95.21%。结论同位素标记示踪法与TCA法结合测定血浆NGF,方法稳定、可靠,灵敏度较高,适于NGF血药浓度的测定。
Objective To establish an isotope tracer precipitation method for determining the concentration of plasma NGF in mice. Methods Radioimmunoassay (~ (125) I) labeled tracer combined with trichloroacetic acid (TCA) precipitation method was used to determine the concentration of NGF. Results The minimum detectable concentration of this method was 2.0 ng / ml, the intra-day and inter-day coefficient of variation (CV) were less than 10%, and the plasma concentration of NGF was in the range of 12.5-400 ng / ml. The correlation coefficient r = 0.9990, the average recovery of NGF in plasma was 95.21%. Conclusion The method of isotope labeled tracer combined with TCA in the determination of plasma NGF is stable, reliable and sensitive. It is suitable for the determination of plasma concentration of NGF.
