目的:研究缺氧性肺动脉高压(HPH)大鼠血浆降钙素基因相关肽(SCGRP)水平,CGRP及碱性成纤维细胞生长因子(bFGF)、血小板源性生长因子(PDGF)在肺内表达和川芎嗪(ligustrazine,Li)对其影响。方法:建立SD大鼠常压HPH模型;用酶联免疫检测技术测定SCGRP水平;用免疫组化染色检测CGRP,PDGF,bFGF在肺内表达水平。结果:缺氧(H)组大鼠SCGRP较正常对照(N)组明显下降,P<0.01;缺氧加川芎嗪(H+Li)组SCGRP水平较H组无显著性差异,P>0.05;免疫组化染色显示H组CGRP,bFGF,PDGF的肺内表达均显著增强,而H+Li组bFGF,PDGF表达水平与H组比较明显降低,但CGRP表达不受Li影响。结论:HPH的形成与SCGRP下降和肺内PDGF,bFGF表达增强有关;Li抑制缺氧大鼠肺内PDGF和bFGF表达是其防治HPH的机制之一。
Objective: To study the plasma levels of calcitonin gene-related peptide (SCGRP) in hypoxic pulmonary hypertension (HPH) rats, and the expression of CGRP, basic fibroblast growth factor (bFGF) and platelet-derived growth factor (PDGF) in the lungs. Ligustriazine (Li) influences it. METHODS: A normal rat model of HPH was established. The level of SCGRP was determined by enzyme-linked immunosorbent assay. The expression of CGRP, PDGF and bFGF in the lung was detected by immunohistochemical staining. RESULTS: SCGRP in the hypoxic (H) group was significantly lower than that in the normal control (N) group, P<0.01; SCGRP levels in the hypoxia plus tetramethylpyrazine (H+Li) group had no significant difference compared with the H group (P> Immunohistochemical staining showed that the expression of CGRP, bFGF and PDGF in the lungs of group H was significantly increased, but the expression of bFGF and PDGF in H+Li group was significantly lower than that in group H, but the expression of CGRP was not affected by Li. Conclusion: The formation of HPH is related to the decrease of SCGRP and the expression of PDGF and bFGF in the lung. Li inhibits the expression of PDGF and bFGF in lungs of hypoxic rats is one of the mechanisms of its prevention and treatment of HPH.
