目的研究嗅三针疗法对AD大鼠学习、记忆功能的改善及其与大脑边缘叶突触素蛋白含量多少的相关性。方法选取成年雄性SD大鼠50只,体重在290±9g。随机每组选取10只大鼠,分为五组即:AD模型组、AD嗅神经切断模型组、实验组、实验对照组及正常对照组。用嗅三针针刺AD大鼠模型及AD嗅神经切断大鼠模型组,针刺结束用水迷宫实验测试实验大鼠学习和记忆能力,采用ELISA法测定大脑边缘叶突触素蛋白含量。结果水迷宫试验结果表明:AD模型组与AD嗅神经切断模型组平均逃避潜伏期和平均游泳路程数据相比,差异无显著性意义(P>0.05);实验对照组小于AD模型组(P<0.05);实验组小于实验对照组(P<0.05);正常对照组和实验组均明显小于AD模型组(P<0.01);大脑边缘叶突触素蛋白含量比较:AD模型组与AD嗅神经切断模型组相比较,差异无显著性意义(P>0.05)。实验组高于实验对照组(P<0.05);正常对照组、实验组和实验对照组均明显高于AD模型组(P<0.01)。结论嗅三针疗法可明显强化AD大鼠学习记忆功能、疗效机制与提高大脑边缘叶突触素蛋白含量有明显相关性。
Objective To study the effect of olfactory three-needle therapy on learning and memory in AD rats and its correlation with the amount of synaptophysin in the marginal lobe of the brain. Methods Fifty adult male Sprague-Dawley rats weighing 290 ± 9g were selected. Ten rats were randomly selected from each group and divided into five groups: AD model group, AD olfactory nerve transection model group, experimental group, experimental control group and normal control group. The rat model of olfactory ensheathing was induced by three acupuncture needle acupuncture and the rat model of AD olfactory nerve transection. The water maze test was used to test the learning and memory abilities of the experimental rats. The content of synaptophysin was measured by ELISA. Results The results of water maze test showed that there was no significant difference between the AD model group and the AD olfactory nerve transection model group (P> 0.05), while the experimental control group was smaller than the AD model group (P <0.05) ); The experimental group was smaller than the experimental control group (P <0.05); The normal control group and the experimental group were significantly smaller than the AD model group (P <0.01); Synaptic protein content in the marginal lobe of the brain: AD model group and AD olfactory nerve cut There was no significant difference between the model group and the model group (P> 0.05). The experimental group was higher than the experimental group (P <0.05); the normal control group, experimental group and experimental control group were significantly higher than the AD model group (P <0.01). Conclusion Olfactory three-needle therapy can significantly enhance learning and memory in AD rats. The therapeutic mechanism is related to the increase of synaptophysin protein content in the marginal zone of the brain.
