用异种或同种鼠胚成纤维细胞和鼠脾细胞共培养研究了日本脑炎病毒(JEV)的潜伏感染。经间接免疫荧光试验(IFA)或小鼠接种证实,用共培养法能从潜伏感染的鼠脾脏T细胞中分离出激活的JEV,但不能从其B细胞和巨噬细胞中分离到。IFA检查表明JEV感染Lyt_1细胞的比例较Lyt_2细胞高;潜伏感染的鼠脾细胞可出现淋巴增殖反应,但比对照鼠要低得多。这些结果提示JEV在T细胞内形成潜伏感染。
Latent infection of the Japanese encephalitis virus (JEV) was studied by co-culturing xenogenic or allogenic mouse embryo fibroblasts with murine splenocytes. Indirect immunofluorescence assay (IFA) or mouse inoculation confirmed that activated JEV was isolated from latent infected murine spleen T cells using co-culture but not isolated from their B cells and macrophages. IFA examination showed that the proportion of Lyt-1 cells infected by JEV was higher than that of Lyt-2 cells; lymphocyte proliferation reaction was observed in the latent infected mouse spleen cells, but much lower than that of the control mice. These results suggest that JEV forms a latent infection in T cells.
