目的探讨柴胡疏肝散对非酒精性脂肪性肝病(NAFLD)大鼠肝组织沉默信息调节因子蛋白1(SIRT1)/p53通路相关蛋白表达的影响。方法将24只雄性SD大鼠随机分为正常对照组、模型组、柴胡疏肝散组(9.6 g·kg-1),每组8只。正常对照组给予基础饲料,模型组和柴胡疏肝散组给予高脂饲料,柴胡疏肝散组同时给予柴胡疏肝散浸膏剂灌胃。16周后处死大鼠,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)含量和肝组织TC、TG含量;取肝组织行HE和油红O染色观察组织学改变;蛋白质印迹法检测肝组织SIRT1、p53、乙酰化p53(Ac-p53)蛋白表达。结果与模型组比较,柴胡疏肝散组肝组织病理学结果表明,脂质沉积明显减轻,血清LDL-C和肝组织TC、TG显著降低(P<0.05,P<0.01),肝组织SIRT1蛋白表达水平显著上调(P<0.01),而Ac-p53蛋白表达水平下调(P<0.05)。结论柴胡疏肝散能够减轻高脂饮食诱导的NAFLD大鼠肝组织脂质沉积,调控肝组织SIRT1/p53通路相关蛋白的表达水平可能是其作用机制之一。
Objective To investigate the effect of Chaihu Shugan Powder on the expression of SIRT1 / p53 pathway-related protein in nonalcoholic fatty liver disease (NAFLD) rats. Methods Twenty-four male SD rats were randomly divided into normal control group, model group and Chaihu Shugan Powder group (9.6 g · kg -1), with 8 rats in each group. The normal control group was given basic diet, model group and Chaihu Shugan Powder group were given high-fat diet, Chaihu Shugan powder group given Chaihu Shugan powder extract at the same time. After 16 weeks, the rats were sacrificed and the levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-C) and low density lipoprotein (LDL- The liver tissue was stained with HE and Oil Red O. The histological changes were observed. The protein expressions of SIRT1, p53 and Ac-p53 in hepatic tissues were detected by Western blotting. Results Compared with the model group, the pathological results of Chaihu Shugan San showed that the lipid deposition was significantly reduced, the levels of serum LDL-C and liver tissue TC and TG were significantly decreased (P <0.05, P <0.01), and the levels of SIRT1 The protein expression level was significantly up-regulated (P <0.01), while Ac-p53 protein expression was down-regulated (P <0.05). Conclusion Chaihu Shugan Powder can reduce the lipid deposition of liver tissue in NAFLD rats induced by high-fat diet and regulate the expression of SIRT1 / p53 pathway-related proteins in liver tissue, which may be one of its mechanisms.
