灌胃番泻叶液建立大鼠腹泻模型,预防实验给药与造模同时进行,共6 d;治疗实验造模12 d,第7天开始给药,给药时间为6 d。实验期间进行症状积分和体重称量,实验结束后摘取心、肝、脾、肾、脑、肾上腺、胸腺称重,并检测血清中乳酸脱氢酶(lactic dehydrogenase,LDH)和心肌肌钙蛋白-I(cardiac troponin-I,c Tn-I)指标。分别研究附子水提物预防和治疗给药对腹泻大鼠的药效及其伴随毒性作用。结果显示附子水提物能改善腹泻大鼠的症状,预防给药药效的半数有效量(ED50)为1.420 4 g·kg-1,治疗给药的药效ED50为1.048 9 g·kg-1。附子水提物同时能降低心体比和升高血清LDH和c Tn-I的水平。得出结论附子水提物对番泻叶致腹泻大鼠具有一定的预防和治疗作用,同时伴有一定的心脏毒性。
The model of diarrhea in rats was established by intragastric administration of senna leaf liquid, and the experimental drug administration and model building were prevented at the same time for 6 days. The treatment experiment was performed on the 12th day and started on the 7th day for 6 days. Symptoms and body weight were weighed during the experiment. Heart, liver, spleen, kidney, brain, adrenal gland and thymus were harvested at the end of the experiment. Serum lactic dehydrogenase (LDH) and cardiac troponin -I (cardiac troponin-I, c Tn-I) index. To study the efficacy and the concomitant toxicity of the aqueous extract of Aconite on prevention and treatment of diarrhea in rats. The results showed that the aqueous extract of Aconite improved the symptoms of diarrhea in rats, and the half-effective dose (ED50) of 1.420 4 g · kg-1 for preventing drug administration was 1.2048 g · kg-1 . The aconite water extract can reduce the body-to-body ratio and increase the levels of serum LDH and cTn-I simultaneously. It was concluded that the water extract of aconite had certain preventive and therapeutic effects on rats with diarrhea caused by senna, accompanied by certain cardiotoxicity.
