目的观察愈灵颗粒对戊四氮(PTZ)致大鼠海马纤维出芽(MFS)的影响。方法选取70只清洁级SD大鼠,从中随机选取10只作为空白对照组。其余动物用戊四氮(PTZ)造模,将符合模型标准的大鼠随机分为3组,即愈灵颗粒组、丙戊酸钠组和模型组。运用Timm染色法观察MFS,探讨愈灵颗粒对海马MFS的影响。结果模型组与空白组比较,大鼠海马IML出芽目测评级差异无统计学意义(P>0.05),海马CA3区神经出芽的黑染点数目和面密度、平均光密度和积分光密度结果比较差异均有统计学意义(P<0.05),综合常规切片结果提示癫可导致海马神经元死亡,进而导致纤维出芽。愈灵颗粒组与模型组比较,愈灵颗粒可降低海马CA3区神经出芽的数目,黑染点数目和面密度、平均光密度和积分光密度结果比较差异均有统计学意义(P<0.01)。结论①致大鼠存在海马MFS现象;②愈灵颗粒可以减少癫大鼠海马MFS的数量和降低阳性程度,说明愈灵颗粒可保护海马神经元,抑制其纤维出芽。
Objective To observe the effect of Yu-Ling-Ling Granule on hippocampal fiber sprouting (MFS) in rats induced by pentylenetetrazole (PTZ). Methods Seventy clean-grade SD rats were selected and randomly selected as blank control group. The remaining animals were treated with pentylenetetrazolium (PTZ), and the rats that meet the model criteria were randomly divided into three groups, namely, Juejiling Granule group, sodium valproate group and model group. Timm staining was used to observe the MFS, to explore the Yu-Ling particles on hippocampal MFS. Results Compared with the blank group, there was no significant difference in visual grading of IML in rat hippocampus (P> 0.05). The number of black spots in hippocampal CA3 area was significantly different from that of the control group in terms of area density, average optical density and integral optical density (P <0.05). Combined with routine slicing results suggest that epilepsy can cause death of hippocampal neurons, leading to fiber sprouting. Compared with the model group, Juelingling granule can reduce the number of neurite outgrowth in CA3 area of hippocampus. The number of black spots and surface density, the average optical density and the integral optical density results were significantly different (P < 0.01). Conclusions ①The hippocampal MFS in induced rats is similar to that in the control rats. ②Yujianling granules can reduce the number of MFS in hippocampus and decrease the positive rate in hippocampus of rats with epilepsy.
