利用电击法将金针茹(Flammulinavelutipes)金杂19菌株的染色体DNA导入金针菇088菌株的单核菌丝原生质体中,根据转化的双核菌丝具“锁状联合”这一形态学特征筛选出了转化菌株。转化率为(7.38~12.5)×10-5,相对转化率为14.0%~18.9%。转化菌株的酯酶同工酶酶谱中出现了亲株没有的新酶带,说明遗传物质发生了变异。出菇实验表明,转化菌株的菌丝生长速度快于金针菇088菌株,现蕾期提前7天左右,菇形较好,生物学效率达91.7%。
The chromosomal DNA of Flammulinavelutipes Jinza 19 was introduced into the protoplasts of mononuclear mycelium of Flammulina velutipes strain 088 by electroporation, and the transformation was screened out according to the morphological characteristics of the “binuclear” Strain. The conversion rate was (7.38 ~ 12.5) × 10-5, the relative conversion rate was 14.0% ~ 18.9%. Esterase isoenzymes in the transformed strains showed no new bands in the parent strain, indicating that the genetic material was mutated. The fruiting experiment showed that the mycelial growth rate of transformed strains was faster than that of Flammulina velutipes 088 strain, the budding stage was about 7 days ahead of schedule, the mushroom shape was better, and the biological efficiency reached 91.7%.
