目的观察大鼠孕期酒精暴露对子代胆碱能神经系统的影响。方法妊娠SD母鼠酒精2g·kg-1·d-1(模型组)或等体积蒸馏水(对照组)灌胃至分娩前。8周龄子代鼠进行水迷宫实验测定学习记忆行为,乙酰胆碱转移酶免疫组化和乙酰胆碱酯酶组织化学检测基底前脑胆碱能神经元和海马胆碱能纤维的变化。结果与对照组相比,模型组学习记忆能力下降(P<0.05),基底前脑胆碱能神经元数目和海马乙酰胆碱能纤维终末减少(P<0.01)。结论孕期酒精暴露可导致子鼠隔-海马胆碱能神经系统损害。
Objective To observe the influence of alcohol exposure during pregnancy on cholinergic neurons in children. Methods Gestational SD rats were administered 2g · kg-1 · d-1 alcohol (model group) or equal volume of distilled water (control group) before gavage. The 8-week-old progeny mice were subjected to water maze test to measure the learning and memory behaviors. The changes of cholinergic neurons and cholinergic fibers in basal forebrain were detected by acetylcholinesterase immunohistochemistry and acetylcholinesterase histochemistry. Results Compared with the control group, the learning and memory ability of the model group decreased (P <0.05), while the number of cholinergic neurons and the number of terminal acetylcholine fibers in the basal forebrain decreased (P <0.01). CONCLUSIONS Alcohol exposure during pregnancy may result in cholinergic neuronal damage in the septum and hippocampus of the offspring.
