目的探讨暖心胶囊对心力衰竭(心衰)模型大鼠的心肌细胞及P53蛋白表达的影响。方法 40只SD大鼠随机分为空白组、模型组、暖心胶囊低剂量组、暖心胶囊高剂量组、地高辛组。除空白对照组外,其余各组均用狭窄腹主动脉方法造模,成功模型灌胃给药,测量心肌重量,观察心肌病理改变,运用免疫组织化学染色法检测P53蛋白。结果与空白组比较,模型组心脏比重明显增加,P53蛋白表达阳性率显著升高,差异有统计学意义(P<0.05);与模型组比较,暖心胶囊低剂量组、暖心胶囊高剂量组、地高辛组能明显降低心脏比重(P<0.05),明显改善心衰大鼠的心肌结构,减少P53表达(P<0.05)。结论暖心胶囊能明显改善心衰大鼠的心肌结构,逆转心肌重构,减少P53表达,拮抗心肌细胞的凋亡。
Objective To investigate the effects of Wenshen Capsule on the expression of P53 protein in cardiomyocytes and heart failure model rats. Methods 40 SD rats were randomly divided into blank group, model group, low-dose warm heart capsule, warm heart capsule high-dose group, digoxin group. Except the blank control group, the other groups were all established by the method of narrow abdominal aorta. The successful model was intragastrically administrated, the myocardial weight was measured, the pathological changes of myocardium were observed, and P53 protein was detected by immunohistochemical staining. Results Compared with the blank group, the cardiac proportion of the model group increased significantly and the positive rate of P53 protein was significantly increased (P <0.05). Compared with the model group, Group, digoxin group can significantly reduce the heart weight (P <0.05), significantly improve myocardial tissue in heart failure rats, reduce P53 expression (P <0.05). Conclusion Warm Heart Capsule can obviously improve myocardial structure, reverse myocardial remodeling, decrease P53 expression and antagonize cardiomyocyte apoptosis in heart failure rats.
