目的比较弗氏完全佐剂(FCA)与单磷脂A(MPL)的免疫增强作用,为提高重组SjGST-Sj32(SjGST-Sj32)的疫苗效果提供依据。方法SjGST-Sj32加弗氏完全佐剂(SjGST-Sj32+FCA)或单磷脂A(SjGST-Sj32+MPL)免疫小鼠3次,ELISA法检测抗体水平,用减虫率及减卵率表示保护性作用。结果与PBS对照组相比,SjGST-Sj32加FCA或MPL,免疫BALBC/c小鼠减虫率分别为48.8%(P<0.01)和44.1%(P<0.01);每克肝卵(LEPG)减少率分别为59.4%(P<0.01)和46.2%(P<0.01);SjGST-Sj32+FCA组抗体滴度达1:204800(P<0.05),而SjGST-Sj32+MPL组抗体滴度为1:51200(P<0.05)。结论FCA和MPL均可以增强对SjGST-Sj32免疫小鼠的抗感染保护性免疫力;FCA也可增强SjGST-Sj32诱导小鼠抗生殖免疫,而MPL似不具明显增强SjGST-Sj32的抗生殖免疫作用。佐剂MPL在提高免疫小鼠体液免疫应答方面稍逊于经典佐剂FCA。
Objective To compare the immunopotentiation effect of Freund’s complete adjuvant (FCA) and monophospholipid A (MPL), and to provide basis for improving the vaccine efficacy of recombinant SjGST-Sj32 (SjGST-Sj32). Methods The mice were immunized three times with SjGST-Sj32 plus complete Freund’s adjuvant (SjGST-Sj32 + FCA) or SjGST-Sj32 + MPL. The antibody levels were detected by ELISA. Results Compared with PBS control group, the worm reduction rates of SjGST-Sj32 plus FCA or MPL in immunized BALBC / c mice were 48.8% (P <0.01) and 44.1% (P <0.01) (P <0.01). The antibody titer in SjGST-Sj32 + FCA group was 1: 204800 (P < 0.05), while the antibody titer in SjGST-Sj32 + MPL group was 1: 51200 (P <0.05). Conclusion Both FCA and MPL can enhance the anti-infective protective immunity against SjGST-Sj32-immunized mice. FCA also enhances the anti-reproductive immunity induced by SjGST-Sj32, while MPL does not significantly enhance the anti-immune effect of SjGST-Sj32 . Adjuvant MPL was slightly inferior to classical adjuvant FCA in enhancing the humoral immune response in immunized mice.
