利用根瘤土壤杆菌Ti质粒转化系统,采用叶盘法将CMV-Cp基因转入番茄细胞中,获得42株转基因植株,经Southernblot和Westernblot检测,证明CMV-cp基因已进入番茄核染色体中,并能正常表达。通过对转基因植株R1和R2代苗期人工接种CMV鉴定,发现CMV-Cp基因产物对CMV有一定抗性,其抗性遗传符合孟德尔一对基因控制性状的遗传规律并能稳定遗传。
Using the Ti plasmid transformation system of Agrobacterium tumefaciens, CMV-Cp gene was transferred into tomato cells by leaf disc method, and 42 transgenic plants were obtained. The result of Southern blot and Western blot showed that CMV-cp gene has entered the chromosome of tomato, Normal expression. CMV-Cp gene products were found to have some resistance to CMV by identifying CMV of transgenic plants R1 and R2 at the seedling stage. The inheritance of CMV-Cp gene was consistent with that of Mendel’s genetic control trait and could be inherited stably.
