目的分析核迁移蛋白细胞核分离基因C(NudC)与肾癌细胞迁移活性的关系。方法采用Transwell方法比较293和OS-RC-2两个细胞株的迁移能力;RT-PCR和Western blot方法分别检测两个细胞株中NudC mRNA和蛋白的表达差异;免疫荧光方法检测NudC蛋白在上述两个细胞株中的定位。结果肾癌细胞株OS-RC-2的迁移能力明显高于胚肾细胞株293。在mRNA水平,人肾癌细胞株OS-RC-2中NudC表达均高于人胚肾细胞株293(2.6±0.3vs.1.0±0.1)(P<0.05)。免疫荧光结果和细胞核蛋白表达结果显示NudC蛋白在人肾癌细胞株OS-RC-2的核内表达高于人胚肾细胞株293(4.1±0.3vs.1.0±0.2)(P<0.05)。结论 NudC蛋白在肾癌细胞株核内的表达高于正常细胞株,提示NudC可能与肾癌细胞迁移活性密切相关。
Objective To analyze the relationship between nuclear transfer protein C (NudC) and the migration of renal cell carcinoma. Methods Transwell method was used to compare the migration ability of 293 and OS-RC-2 cell lines. The expression of NudC mRNA and protein was detected by RT-PCR and Western blot respectively. Immunofluorescence method was used to detect the expression of NudC protein in the above-mentioned Positioning in two cell lines. Results The migration ability of renal cell carcinoma cell line OS-RC-2 was significantly higher than that of embryonic kidney cell line 293. At the mRNA level, the expression of NudC in human renal cell carcinoma cell line OS-RC-2 was higher than that in human embryonic kidney cell line 293 (2.6 ± 0.3 vs.1 ± 0.1) (P <0.05). The results of immunofluorescence and nuclear protein expression showed that the expression of NudC protein in human renal cell carcinoma cell line OS-RC-2 was higher than that of human embryonic kidney cell line 293 (4.1 ± 0.3 vs.1 ± 0.2) (P <0.05). Conclusion The expression of NudC protein in the nucleus of renal cell carcinoma is higher than that in normal cell lines, suggesting that NudC may be closely related to the migration of renal cell carcinoma.
