观察到环肽类抗生素多粘菌素B对小鼠肉瘤S180细胞增殖具有显著抑制作用,半数有效抑制浓度(IC50)为8.95×105IU·L-1。用毛细管柱气相色谱法分析了多粘菌素B作用于S180细胞24h后膜脂肪酸组成的变化,发现磷脂的花生四烯酸(C20:4)显著减少,中性脂的硬脂酸(C18:0)增多,油酸(C18:1)减少,硬脂酸与油酸的比值(C18:0/C18:1)升高.提示多粘菌素B对S180细胞增殖的抑制与膜脂肪酸组成的变化有关。
Cyclic antibiotic polymyxin B was found to inhibit the proliferation of murine sarcoma S180 cells significantly. The IC50 value was 8.95 × 105 IU · L-1. The changes of membrane fatty acid composition of S180 cells treated with polymyxin B were analyzed by capillary gas chromatography. The results showed that the content of arachidonic acid (C20: 4) of phospholipids was significantly decreased, the content of stearic acid (C18: 0) increased, oleic acid (C18: 1) decreased, stearic acid and oleic acid ratio (C18: 0 / C18: 1) increased.
