目的观察玻璃苣醇提取物对去势小鼠免疫功能及脑内神经递质的影响,探讨玻璃苣对更年期小鼠模型的作用及其作用机制。方法建立小鼠更年期模型,测定小鼠的脂肪系数和Less指数以及脾系数和胸腺系数;采用酶联免疫吸附法(ELISA)检测各组小鼠脑组织匀浆液中多巴胺(DA)、去甲肾上腺素(NA)、5-羟色胺(5-HT)的含量。结果玻璃苣醇提取物对脂肪系数和Less指数无明显的影响,能显著升高脾系数和胸腺系数;各组均有升高去势小鼠脑内DA,NA,降低去势小鼠脑内5-HT含量的作用,其中玻璃苣醇提取物高剂量组、中剂量组及更年安组差异均有统计意义(P<0.05或P<0.01)。结论玻璃苣醇提取物能明显增加更年期去势小鼠的脾系数和胸腺系数,对免疫系统有一定的保护作用;玻璃苣醇提取物在一定程度上能升高去势小鼠脑内降低的DA和NA含量,降低去势小鼠脑内异常升高的5-HT含量。
Objective To observe the effect of the extract of Echinoccerus on immunological function and brain neurotransmitter in ovariectomized mice and to explore the role of borage in climacteric mouse model and its mechanism. Methods Mouse model of climacteric period was established, and the mice were assessed for fat index, Less index and spleen index and thymus index. The levels of dopamine (DA), noradrenal gland (NA), serotonin (5-HT) content. Results The results showed that there was no significant effect of the extract of Cichorium intimae on the fat index and Less index, and the spleen index and thymus index were significantly increased. DA and NA in brains of sham-operated mice increased (P <0.05 or P <0.01). There was significant difference between the high dose group, the medium dose group and the senilean group (P <0.05 or P <0.01). Conclusion The extract of chicory can significantly increase the spleen coefficient and thymus coefficient of clonergic mice with castrated ovaries, which may have some protective effect on the immune system. The extract of chicory alcohol could elevate the decreased brain in castrated mice to some extent DA and NA content, reduce the abnormally elevated 5-HT content in the brain of castrated mice.
