按Selye法结扎Wistar大鼠冠状动脉(简称冠脉)左前降支复制早期心肌缺血模型;又经颈外静脉注射垂体后叶素复制冠脉痉挛模型。按心肌不同缺血时间(25min,1h,3h)及对照组将以上各模型均分成4组。每组心肌取心尖及其相邻组织块制成石蜡切片,进行HE染色、链霉菌素一生物素肌红蛋白免疫组织化学染色(LSAB-Mb染色)。结果表明:早期心肌缺血组心肌呈片状肌红蛋白(Mb)缺失;冠脉痉挛组呈多发性、小灶性缺失。所以,LSAB-Mb法可望为冠脉痉挛所致的心肌缺血提供客观形态学证据。
According to Selye method, Wistar rat coronary artery (referred to as the coronary artery) left anterior descending artery branch early myocardial ischemia model; and intravenous injection of vasopressin copy coronary artery spasm model. According to different myocardial ischemic time (25min, 1h, 3h) and control group, the above models were divided into 4 groups. Each group of cardiac apex and its adjacent tissue made of paraffin sections, HE staining, streptomycin-biotin myoglobin immunohistochemical staining (LSAB-Mb staining). The results showed that there was a lack of Mb in the early myocardial ischemia group. The coronary spasm group showed multiple and focal lesion. Therefore, LSAB-Mb method is expected to provide objective morphological evidence of myocardial ischemia caused by coronary spasm.
