目的:分析异钩藤碱在大鼠体内的代谢途径及产物。方法:异钩藤碱按剂量37.5 mg·kg-1给药后,收集血浆,在正离子模式下,采用UPLC-Q-TOF-MS分析异钩藤碱在大鼠体内的代谢途径,流动相0.1%甲酸水溶液-乙腈梯度洗脱。结果:通过与空白生物样品和对照品的提取离子色谱图、质谱碎片裂解信息比对,共鉴别出13个成分,其中1个原型,12个代谢产物,包括Ⅰ相代谢产物11个,Ⅱ相代谢产物1个。结论:异钩藤碱在大鼠体内的代谢途径为氧化、立体异构、羟基化、脱氢、水解及葡萄糖醛酸化等,丰富了吲哚类生物碱的ESI-MS裂解规律,为此类化合物的质谱解析提供参考。
OBJECTIVE: To analyze the metabolic pathway and products of isorhline in rats. Methods: The plasma was collected after the administration of isoniastatine at a dose of 37.5 mg · kg-1.Under the positive ion mode, the metabolic pathway of isorhline in rats was analyzed by UPLC-Q-TOF-MS. The mobile phase 0.1% formic acid in water - acetonitrile gradient. Results: 13 components were identified by extraction ion chromatogram and mass fragment fragmentation information of blank biological samples and reference materials, including 1 prototype, 12 metabolites, including 11 phase Ⅰ metabolites, Ⅱ phase 1 metabolite. CONCLUSION: The metabolic pathway of isorhynchophylline in rats is oxidation, stereoisomerism, hydroxylation, dehydrogenation, hydrolysis and glucuronidation, which enriches the ESI-MS cleavage of indole alkaloids. Reference for mass spectrometry analysis of compounds.
