目的通过分析HEV的5’-UTR基因序列,确定上海市徐汇区手足口病(HFMD)病例中HEV的型别。方法采集2012年HFMD患儿标本,采用Real time-RT-PCR分别检测HEV、EV71和CoxA16核酸,初步鉴定其型别。从中挑选未能分型的HEV毒株扩增其5’-UTR进行核苷酸序列测定,运用DNAStar和MEGA5.2软件进行序列分析。结果26例标本均为HEV阳性:10例EV71阳性,5例CoxA16阳性,11例其他型HEV阳性。扩增11例未能分型的HEV标本的5’-UTR序列,9例测序成功,通过序列比对确定病毒型别:CoxA16 8例,CoxA10 1例。结论上海市徐汇区HFMD患儿的主要病原是EV71和CoxA16,也存在着其他型别的HEV,如CoxA10,应加强HEV基因亚型的分布及流行特点的监测。
Objective To determine the type of HEV in HFMD in Xuhui District, Shanghai by analyzing the sequence of 5’-UTR of HEV. Methods Children with HFMD were collected from 2012, and HEV, EV71 and CoxA16 were detected by Real time-RT-PCR, respectively. The unselected HEV strains were selected to amplify their 5’-UTR for nucleotide sequence determination, and DNAStar and MEGA5.2 software were used for sequence analysis. Results All the 26 specimens were positive for HEV: 10 EV71 positive, 5 CoxA16 positive and 11 other HEV positive. The sequence of 5’-UTR of 11 untreated HEV specimens was amplified. Nine of them were sequenced successfully. The virus types were identified by sequence alignment: CoxA16 in 8 cases and CoxA10 in 1 case. Conclusion The major pathogens of HFMD in Xuhui District of Shanghai are EV71 and CoxA16, and other types of HEV, such as CoxA10, should be strengthened to monitor the distribution and prevalence of HEV genotypes.
