目的采用HPLC法测定小鼠脑匀浆中的盐酸尼莫司汀。方法使用Diamonsil C18柱,流动相为甲醇-水-三乙胺(20∶79.5∶0.5,磷酸调pH3),流速1.0mL·min-1,检测波长241nm,柱温35℃。结果盐酸尼莫司汀0.20~5.00mg·L-1与峰面积的线性关系良好(r=0.9999);平均方法回收率95.7%,平均萃取回收率101.2%,日内、日间RSD均<5%。结论该方法简便、灵敏、准确,可用于测定脑匀浆中的盐酸尼莫司汀。
Objective To determine nimustine hydrochloride in mouse brain homogenate by HPLC. Methods Diamonsil C18 column was used. The mobile phase was methanol-water-triethylamine (20:79.5:0.5, adjusted to pH3 with phosphoric acid) and the flow rate was 1.0 mL · min-1. The detection wavelength was 241 nm and the column temperature was 35 ℃. Results The linear relationship between nimodipine hydrochloride 0.20 ~ 5.00mg · L-1 and peak area was good (r = 0.9999). The average recovery rate was 95.7% and the average extraction recovery rate was 101.2% . Conclusion The method is simple, sensitive and accurate and can be used for the determination of nimodipine hydrochloride in brain homogenates.
