TTC在螺旋平板法测定冷冻水产品中菌落总数的应用研究

研究以2,3,5-三苯基氯化四氮唑(TTC)作为指示剂在螺旋平板法测定冷冻水产品中菌落总数的适宜用量。通过对大肠埃希氏菌、金黄色葡萄球菌、枯草芽孢杆菌三株标准菌株制备的菌悬液在含梯度含量TTC的平板计数琼脂(PCA)中分别进行菌落计数、生长和显色观察实验,初步筛选出适宜的TTC含量范围。以冷冻水产品样品在该用量范围内的菌落计数和显色观察实验,验证在实际应用中的适宜性。验证实验结果显示:在TTC含量为1.5mg/100ml的组与未添加TTC的对照组菌落计数结果经统计学检验无显著性差异,且菌落显色情况良好,有利于计数的准确性。 The appropriate amount of colony in chilled aquatic products was determined by spiral plate method with 2,3,5-triphenyltetrazolium tetrazolium (TTC) as an indicator. The colony count, growth and color observation were carried out on the plate-counting agar (PCA) containing gradient content TTC by using bacterial suspension prepared from three standard strains of Escherichia coli, Staphylococcus aureus and Bacillus subtilis, Preliminary screening of the appropriate range of TTC content. The colony counting and color observation experiments of chilled aquatic product samples within the dosage range were carried out to verify the suitability in practical application. The validation results showed that there was no significant difference in the colony counting results between the groups with the TTC content of 1.5 mg / 100 ml and the control group without the addition of TTC, and the coloration of the colony was good, which is in favor of the accuracy of the counting.
